  • Genetic diversity of Bassia scoparia (L.) A.J. Scott population revealed by using RAPD markers in Dorud, Iran

    The genetic diversity of One hundred and twenty-five feral populations of Bassia scoparia (L.) A.J. Scott collected from twenty isolated locations in Dorud was studied by using eighteen randomly amplified polymorphic DNA. DNA extraction from young and fresh leaves of plants was isolated using CTAB technique. Matrix 1/0 bands RAPD by MVSP Ver 3.2 and NTSYSpc Ver 2.02 clustering UPGMA of Nei and Li coefficient and similar groupings were obvious in the PCoA analysis. A total of eighteen RAPD primers produced of reproducible bands which 13085 produced which 9436 of them were polymorphic (72.11%). The total length of bands created between 115(Q-11) to 1700(Q-07) bp fragment was variable. The results showed the presence of five separated genetic group among studied populations over Dorud. The highest genetic similarity coefficient (0.97) was observed between Ds9 and Ds16 and lowest similarity was between Ds4 and Ds13 (0.40). There was no significant relationship observed between the genetic distance of populations and their geographic distribution. Overall, Randomly Amplified Polymorphic DNA technique could be used for genetic differentiation of diverse populations of Bassia scoparia (L.) A.J. Scott to provide information on their population genetic structure.
